Total Phenolic and Flavonoid Content and In Vitro Antioxidant Activity of Selected Herbal Products Using Oxygen Radical Absorbance Capacity, Multi-radical (ORAC MR5) Assays

Reactive oxygen and nitrogen species (ROS/RNS) are known to cause oxidative damage cells macromolecules. Intake of antioxidant-rich fruits vegetables has been touted as a strategy mitigate stress, but this is not always accommodated in modern diets. Augmentation with herbal supplements may be preventive measure against the negative effects ROS/RNS. In study, we verify compare antioxidant claims by measuring their phytochemical content capacities through radical absorption capacity–multi-radical (ORAC MR5) assays. ORAC MR5 measures activity five biologically relevant ROS/RNS (peroxyl hydroxyl radical, superoxide anion, singlet oxygen, peroxynitrite). Four products were tested: two multi-herbal capsules (INT Oxxy), mangosteen fruit pulp powder capsule (MPC), mixed drink (MHD). Phytochemical was quantified total phenolic flavonoid Oxxy had highest per serving (106.4 mg catechin eq/ 450 capsule) value (37,503 μmol Trolox capsule), followed INT (53.5 eq 17,298 200 MPC (48.1 7,675 500 then MHD (7.4 146.7 30 mL drink). Significant activities peroxyl radicals observed for Oxxy, whereas mostly towards radicals. A strong correlation found between contents values, confirmed supplier on standardized or Oxxy.
